Rofecoxib was approved by the FDA to treat osteoarthritis, rheumatoid arthritis, juvenile rheumatoid arthritis, acute pain conditions, migraine, and dysmenorrhea. The metabolic products are cis-dihydro and trans-dihydro derivatives of rofecoxib which are primarily excreted through urine. Rofecoxib crossed the placenta and blood–brain barrier, and took 1–3 hours to reach peak plasma concentration with an effective half-life (based on steady-state levels) around 17 hours. The therapeutic recommended dosages were 12.5, 25, and 50 mg with an approximate bioavailability of 93%. This was largely based on the VIGOR (Vioxx GI Outcomes Research) study, which compared the efficacy and adverse effect profiles of rofecoxib and naproxen. Īt the time of its withdrawal, rofecoxib was the only coxib approved in the United States with clinical evidence of its superior gastrointestinal adverse effect profile over conventional NSAIDs. Though the class of coxibs includes several agents, degrees of COX-2 selectivity vary among them, with celecoxib (Celebrex) being the least COX-2 selective, and rofecoxib (Vioxx), valdecoxib (Bextra), and etoricoxib (Arcoxia), being highly COX-2 selective. Rofecoxib is a selective COX-2 inhibitor, or "coxib". By creating "selective" NSAIDs that inhibit COX-2, but not COX-1, the same pain relief as traditional NSAIDs is offered, but with greatly reduced risk of fatal or debilitating peptic ulcers. COX-1 mediates the synthesis of prostaglandins responsible for protection of the stomach lining, while COX-2 mediates the synthesis of prostaglandins responsible for pain and inflammation. Ĭyclooxygenase (COX) has two well-studied isoforms, called COX-1 and COX-2. Traditional NSAIDs are avoided in this population due to their effects on platelet aggregation and risk of gastrointestinal ulcers, and high potency opioids are the current standard of care in treating HA. It is the largest cause of morbidity in patients with hemophilia and has no currently approved treatment options in the United States. HA is a degenerative joint disease caused by recurrent intra-articular bleeding. Tremeau announced that the FDA had granted an orphan designation for TRM-201 (rofecoxib) for the treatment of HA, and that they had received FDA feedback on their development plan. In November 2017, Massachusetts-based Tremeau Pharmaceuticals announced its plan to return rofecoxib (TRM-201) to market as a treatment for hemophilic arthropathy (HA). Based on data up to 2015, the FDA reasserted the likelihood of an increased risk of serious adverse CV events from COX-2–selective and nonselective NSAIDs, dependent on dose and duration. In 2005, the FDA issued a memorandum concluding that risks for serious cardiovascular (CV) events seem to be as great for nonselective NSAIDs as for COX-2–selective agents such as rofecoxib, according to long-term, controlled clinical trials. In the year before withdrawal, Merck had sales revenue of US$2.5 billion from Vioxx. Rofecoxib was one of the most widely used drugs ever to be withdrawn from the market. Merck withdrew the drug after disclosures that it withheld information about rofecoxib's risks from doctors and patients for over five years, allegedly resulting in between 88,000 and 140,000 cases of serious heart disease. In September 2004, Merck voluntarily withdrew rofecoxib from the market because of concerns about increased risk of heart attack and stroke associated with long-term, high-dosage use. Worldwide, over 80 million people were prescribed rofecoxib at some time. Rofecoxib gained widespread use among physicians treating patients with arthritis and other conditions causing chronic or acute pain. Rofecoxib was available by prescription in both tablets and as an oral suspension. Rofecoxib was approved in the US by the US Food and Drug Administration (FDA) in May 1999, and was marketed under the brand names Vioxx, Ceoxx, and Ceeoxx. to treat osteoarthritis, rheumatoid arthritis, juvenile rheumatoid arthritis, acute pain conditions, migraine, and dysmenorrhea. Rofecoxib is a COX-2-selective nonsteroidal anti-inflammatory drug ( NSAID). 1/1/2024 0 Comments Jumex pear nectarBe sure to stock them all to appeal to every guest. Everyone has a liquor they can’t drink anymore and one that serves as their go-to spirit. My years as a bartender taught me a lesson about what people are willing to drink. Every stocked home bar needs the usual well liquors: vodka, rum, whiskey, gin, and if you’re brave…tequila. Unflavored vodka isn’t sweet and goes well with just about any flavor. Sometimes a holiday inspires my cocktail recipes but other times it’s just the discovery of a flavored water, juice, or soda I haven’t tried yet. Boo-rific Blood Orange Cocktail is made even better by adding flavored vodka. You don’t have time to prep sangria fruit or brainstorm a punch bowl recipe. Some of you are busy this time of year balancing trick or treat night with the kids, classroom Halloween parties, and hey…maybe your own spirited affair just for adults. What’s great about these drinks is that they are super simple to make for party guests. Since I dress up myself and my house, of course I would dress up my Halloween themed cocktails. We won Best Costumed Couple last year as the beekeeper, queen bear, and baby bee (our dog) so the expectation for our costumes this year is intense. Of course, this year’s costume is super secret until Halloween night. Shout out to mom who set the bar high by sewing or helping me create my costumes from a young age. But you know what? The fang-tastic holiday will remain one of my favorite celebrations dating all the way back to my first clown costume in the kindergarten classroom party. I’ve shown up at costume parties as Amy Winehouse, a 20s flapper, zombie prom queen, scarecrow, 80s chick, and queen bee. Halloween is my excuse to dress up in costume. Give the Peaty Pera a try on one of these cold winter’s nights.Once every year I become someone I’m not. So we added a bit of thyme simple syrup (you can find a recipe for that in our Thyme to Shine cocktail) which really worked well with the flavors of pear and the amaro. Not wanting to just add sweetness, we decided to layer in a little more flavor. The drink was a little tart and we needed to bring the drink into balance with some added sweetness. We chose to use Amaro Lucano, an Italian amaro that has a nice blend of herbs and spices, with a great finish of warm baking spice and hint of bitterness.Ī splash of fresh squeezed lemon adds a bright citrus note that brings out the fruity notes of pear. Likewise, one way to bring some rich spicy complexity is to mix in a little amaro. If you want to learn more about Scottish Whisky, make sure to read our Scotch Regions guide!Ī good winter drink needs to have a nice layer of spice. This flavor is reminiscent of a roaring fire on the hearth which really works in a winter drink. We thought pairing it up with some pear nectar made perfect sense. The smoky flavors from the peat used to roast the malt imparts a deep dark flavor in the Scotch. Scotches have a rich malted flavor that works really well with sweet fruits like ripe pears and berries. Thyme Simple Syrup (instructions below!). Mixing Glass / Bar Spoon What Ingredients are Needed for the Peaty Pera? The homemade pear nectar was naturally sweet tart with a bright pear note and body that adds a decadent smoothness to a cocktail. We had made some homemade pear nectar for our Pink Pear cocktail so with that handy, it was only natural to use in the Peaty Pera. Since we have these great pears in season now, we figured a cocktail that combined two of our favorite winter flavors would be a real winner. This is why they start appearing in the dead of winter! Traditional varieties like the Anjou or the Bosc actually need to be exposed to temps below freezing for 4-6 weeks to fully ripen. So to fully allow the fruit to ripen, they need to be exposed to cold to prevent the pear from rotting. Pears, unlike apples, ripen from the inside out. Most people don’t realize that many of the great pear varieties like the Anjou or Bosc actually need to be chilled to ripen due to the unique way that pears ripen.Ĭoupe / Jigger Why are Pears Ripe in the Winter? We recently have been getting in some really great pears as the winter pears are finally fully ripe and ready to go. Middle Tennessee comes into Thursday's contest in a similar fashion to the 49ers, having lost each of its final two regular season games on the road to North Texas and at UTEP. That league tournament run was ended before it began due to the outbreak of COVID-19 the day of the Niners potential game with then-fifth-seeded FIU. Charlotte ended the regular season with wins in four of its last six games of the regular season, earning the fifth seed in Conference USA, which is the program's highest seed in the league tournament since finishing fourth in 2019-20. Junior Brice Williams was named to the All-League Second Team, while sophomores Lu'Cye Patterson and Aly Khalifa earned Honorable Mention acclaim. CLT saw a trio of players earn All-Conference Accolades the league announced Tuesday afternoon. The last time CLT advanced to the semifinals of a conference tournament came in 2008 when the Niners fell to Temple in the semis of the A-10 Championship. A win would also push Charlotte to the semifinals of the C-USA Tournament for the first time since the program rejoined the league in the Fall of 2013. A win would be CLT's first conference tournament victory since the aforementioned 2016 C-USA Tournament when the Niners took down Rice, 79-69 in the opening round of that year's tournament. This contest will be Charlotte's first appearance in the quarterfinal round of the C-USA Tournament since falling to Middle Tennessee in 2015-16 at Legacy Arena in Birmingham, Alabama. Thursday's C-USA Tournament bout will be the 18th all-time meeting between the 49ers and Blue Raiders on the collegiate hardwood in a series dating to 1988, with this being the fourth contest on a neutral floor. All will have an app that displays the collected fitness information and connects users to the broader community. Most will also track a vast selection of sports, either automatically or otherwise. What features should I look out for?Īll of the items on this list will track the same set of basic fitness metrics: steps taken, distance travelled and calories burned. All of the major manufacturers (Fitbit, Garmin, Huawei, Xiaomi and so on) sell fitness trackers that cater to the top and bottom ends of the £100 bracket. The expensive entries are very nearly full-blooded smartwatches but, ordinarily, you should be able to find a cracking fitness tracker for less than £100 or even £50 if your needs are modest. The fitness trackers on this list range in price from around £30 to almost £200. They will often track basic fitness information (steps taken, calories burned, distance travelled) onboard, but tend to require a permanent connection to a smartphone for in-depth analysis, GPS tracking and to display notifications. On the flip side, most fitness trackers are simple bands, with tiny displays. They will pack a combination of large watch faces, app support, Wi-Fi, NFC, Bluetooth, built-in GPS and many, many other jazzy features. They will allow you to monitor pretty much any physical activity, and will most likely keep track of your heart rate and sleep patterns at the very least. These days, almost all smartwatches – from the Apple Watch Series 8 or Samsung Galaxy Watch 5 – are also accomplished fitness trackers. As a raw material producer that transformed itself into a battery manufacturer, Ganfeng Lithium started construction of a 5.4 billion yuan ($758.61 million) solid-state battery production base in Chongqing in July 2022. Over the last couple of years, Ganfeng Lithium was one of the companies leading a new trend of vertical integration in China’s lithium battery industry, in which electric battery companies are investing in raw materials, and raw material companies are producing batteries. The company claims that its solid-state battery is independently developed, including high-grade lithium sulfide raw materials for a flexible electrolyte membrane, making the battery more efficient and safe, as well as with a longer life cycle. Ganfeng Lithium is the very first company, however, to announce the commencement of mass production. But CATL has not yet commenced mass production of condensed batteries or solid-state batteries.Ī range of Chinese and foreign companies have for years made pronouncements on solid-state batteries: Volkswagen and Nissan, for example, plan to launch mass production around 2025. In April 2023, CATL, China’s battery king, announced a new type of battery, the condensed battery, which the company claims can reach an energy density of up to 500 Wh/kg. This is compared to the theoretical threshold of 350 Wh/kg for mainstream lithium-ion batteries that still power almost all electric vehicles (EVs) on the road today, although most of these batteries have an energy density of just over 200 Wh/kg. In addition, CATL has built the world’s largest after-sales service network in this field, establishing 81 cooperation stations, six regional warehouses, four maintenance centers and three training centers in 42 countries including Europe, North America, Asia and Australia.Ganfeng Lithium’s first generation battery will have an energy density of 260 watt-hour per kilogram (Wh/kg), and the second generation will be able to reach an energy density of 400 Wh/kg. Up to now, CATL batteries have been sold in 56 countries and regions around the world, with a cumulative load of more than 5 million vehicles. In terms of manufacturing, CATL has built the largest 5G enterprise private network in China, which has now exceeded 5 million square meters. On August 28, Robin Zeng said at the WNEVC 2022 that full-scale electrification also requires strong manufacturing and delivery capabilities as support. CATL revealed that the Kirin battery will be mass-produced and released in 2023. This system integration reaches a global high, the volume utilization rate exceeds 72%, and the energy density can reach 255Wh/kg, realizing a vehicle range of 1,000 km. On June 23, CATL released its CTP3.0 Kirin battery. Zeng has also said that the combination for innovation of power battery chemical systems, innovation of structures, innovation of manufacturing, plus innovation of business models can promote the development of the new energy vehicle industry. Robin Zeng, the chairman of CATL, once said that besides the Kirin battery, CATL is tackling more new batteries, such as all-solid-state batteries, semisolid-state batteries and condensed matter batteries. In short, these batteries are more stable, with greater capacity and faster conduction efficiency. Some experts say that the core of the condensed state battery is the ability to achieve a superfluid state, superconducting state, stability, and superior conduction.
